Pathogenic — the classification assigned by GeneDx to NM_000117.3(EMD):c.103G>T (p.Glu35Ter), citing GeneDx Variant Classification (06012015). This variant lies in the EMD gene (transcript NM_000117.3) at coding-DNA position 103, where G is replaced by T; at the protein level this means converts the codon for glutamic acid at residue 35 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: The E35X pathogenic variant in the EMD gene has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. This variant is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. The E35X variant was not observed in approximately 6300 individuals of European and African American ancestry in the NHLBI Exome Sequencing Project, indicating it is not a common benign variant in these populations. We interpret E35X as a pathogenic variant
