NM_001356.5(DDX3X):c.80dup (p.Ser28fs) was classified as Pathogenic by GeneDx, citing GeneDx Variant Classification (06012015): The c.80dupA variant in the DDX3X gene has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. The c.80dupA variant causes a frameshift starting with codon Serine 28, changes this amino acid to a Glutamic Acid residue, and creates a premature Stop codon at position 23 of the new reading frame, denoted p.Ser28GlufsX23. This variant is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. The c.80dupA variant was not observed in approximately 6000 individuals of European and African American ancestry in the NHLBI Exome Sequencing Project, indicating it is not a common benign variant in these populations. We interpret c.80dupA as a pathogenic variant.
