NM_006766.5(KAT6A):c.3432dup (p.Pro1145fs) was classified as Pathogenic by GeneDx, citing GeneDx Variant Classification (06012015). This variant lies in the KAT6A gene (transcript NM_006766.5) at coding-DNA position 3432, duplicating one base; at the protein level this means shifts the reading frame starting at proline residue 1145, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: The c.3432dupA pathogenic variant in the KAT6A gene has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. The c.3432dupA variant causes a frameshift starting with codon Proline 1145, changes this amino acid to a Threonine residue, and creates a premature Stop codon at position 30 of the new reading frame, denoted p.Pro1145ThrfsX30. This variant is predicted to cause loss of normal protein function through protein truncation. The c.3432dupA variant was not observed in approximately 6,500 individuals of European and African American ancestry in the NHLBI Exome Sequencing Project, indicating it is not a common benign variant in these populations. We interpret c.3432dupA as a pathogenic variant.
