Pathogenic — the classification assigned by GeneDx to NM_000444.6(PHEX):c.508_529del (p.Leu170fs), citing GeneDx Variant Classification (06012015): The c.508_529del22 pathogenic variant in the PHEX gene causes a frameshift starting with codon Leucine 170, changes this amino acid to a Lysine residue and creates a premature Stop codon at position 44 of the new reading frame, denoted p.Leu170LysfsX44. This pathogenic variant is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. The variant was not observed in approximately 6,500 individuals of European and African American ancestry in the NHLBI Exome Sequencing Project, indicating it is not a common benign variant in these populations.
