This variant lies in the CYP7B1 gene (transcript NM_004820.5) at coding-DNA position 793, where C is replaced by T; at the protein level this means converts the codon for glutamine at residue 265 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: The Q265X pathogenic variant in CYP7B1 is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. It was not observed in approximately 6,500 individuals of European and African American ancestry in the NHLBI Exome Sequencing Project, indicating it is not a common benign variant in these populations. Although Q265X has not been reported previously to our knowledge, we consider it to be pathogenic.
