NM_015338.6(ASXL1):c.3806C>G (p.Ser1269Trp) was classified as Uncertain significance for Inborn genetic diseases by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.S1269W variant (also known as c.3806C>G), located in coding exon 13 of the ASXL1 gene, results from a C to G substitution at nucleotide position 3806. The serine at codon 1269 is replaced by tryptophan, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
