NM_005188.4(CBL):c.1942A>C (p.Ser648Arg) was classified as Uncertain significance by GeneDx, citing GeneDx Variant Classification (06012015). This variant lies in the CBL gene (transcript NM_005188.4) at coding-DNA position 1942, where A is replaced by C; at the protein level this means replaces serine at residue 648 with arginine — a missense variant. Submitter rationale: The S648R variant in the CBL gene has not been reported previously as a pathogenic variant, nor as a benign variant, to our knowledge. The S648R variant is observed in 22/24,026 (0.09%) alleles from individuals of African background in large population cohorts (Lek et al., 2016). The S648R variant is a semi-conservative amino acid substitution, which may impact secondary protein structure as these residues differ in some properties. However, in-silico analyses, including protein predictors and evolutionary conservation, support that this variant does not alter protein structure/function. We interpret S648R as a variant of uncertain significance.
