NM_001367721.1(CASK):c.523dup (p.Val175fs) was classified as Pathogenic by GeneDx, citing GeneDx Variant Classification (06012015): The c.523dupG variant in the CASK gene causes a frameshift starting with codon Valine 175, changes this amino acid to a Glycine residue and creates a premature Stop codon at position 54 of the new reading frame, denoted p.V175GfsX54. This variant is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. It was not observed in approximately 6,500 individuals of European and African American ancestry in the NHLBI Exome Sequencing Project, indicating it is not a common benign variant in these populations. Although the c.523dupG variant has not been previously reported to our knowledge, we interpret this variant to be pathogenic.
