Pathogenic for Psoriasiform dermatitis; Pustule; Erythematous plaque; Netherton syndrome — the classification assigned by 3billion to NM_006846.4(SPINK5):c.1302+4A>T, citing ACMG Guidelines, 2015: The variant is observed at an extremely low frequency in the gnomAD v2.1.1 dataset (total allele frequency: 0.002%). Functional studies provide supporting evidence of the variant having a damaging effect on the gene or gene product (PMID: 25710899). The variant has been reported to be in trans with a pathogenic variant as either compound heterozygous or homozygous in at least 2 similarly affected unrelated individuals (PMID: 16601670, 25710899) and to co-segregate with the disease in at least one similarly affected relative/individual in the same family or similarly affected unrelated family (PMID: 25710899). The variant has been reported at least twice as pathogenic with clinical assertions and evidence for the classification (ClinVar ID: VCV000279898). Therefore, this variant is classified as pathogenic according to the recommendation of ACMG/AMP guideline.
