Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000478.6(ALPL):c.984C>G (p.Phe328Leu), citing Invitae Variant Classification Sherloc (09022015): This variant is not present in population databases (gnomAD no frequency). This sequence change replaces phenylalanine, which is neutral and non-polar, with leucine, which is neutral and non-polar, at codon 328 of the ALPL protein (p.Phe328Leu). This missense change has been observed in individual(s) with hypophosphatasia (PMID: 31707452). This variant is also known as TNSALP F311L. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt ALPL protein function. Experimental studies have shown that this missense change affects ALPL function (PMID: 15660230).
