Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_033026.6(PCLO):c.2644C>T (p.Arg882Ter), citing Invitae Variant Classification Sherloc (09022015): This sequence change creates a premature translational stop signal (p.Arg882*) in the PCLO gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in PCLO are known to be pathogenic (PMID: 25832664, 30287594). For these reasons, this variant has been classified as Pathogenic. This variant has not been reported in the literature in individuals affected with PCLO-related conditions. This variant is not present in population databases (gnomAD no frequency).
