NM_031443.4(CCM2):c.214C>T (p.Gln72Ter) was classified as Pathogenic by GeneDx, citing GeneDx Variant Classification (06012015): The Q72X nonsense variant in the CCM2 gene is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. The Q72X variant was not observed in approximately 6,500 individuals of European and African American ancestry in the NHLBI Exome Sequencing Project, indicating it is not a common benign variant in these populations. Given the available evidence, we interpret Q72X as a pathogenic variant.
