This variant lies in the GCDH gene (transcript NM_000159.4) at coding-DNA position 1186, where G is replaced by A; at the protein level this means replaces aspartic acid at residue 396 with asparagine — a missense variant. Submitter rationale: This sequence change replaces aspartic acid, which is acidic and polar, with asparagine, which is neutral and polar, at codon 396 of the GCDH protein (p.Asp396Asn). This missense change has been observed in individual(s) with clinical features of glutaric acidemia type I (Invitae). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. This variant is not present in population databases (gnomAD no frequency).
