NM_000535.7(PMS2):c.270T>C (p.Ser90=) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The c.270T>C variant (also known as p.S90S), located in coding exon 4 of the PMS2 gene, results from a T to C substitution at nucleotide position 270. This nucleotide substitution does not change the amino acid at codon 90. This nucleotide position is not well conserved in available vertebrate species. In silico splice site analysis for this alteration is inconclusive; however, direct evidence is insufficient at this time (Ambry internal data). Based on the available evidence, the clinical significance of this variant remains unclear.
