To our knowledge, no occurrence of c.580C>T in individuals affected with Metachromatic Leukodystrophy and no experimental evidence demonstrating its impact on protein function have been reported. Different variants affecting the same codon have been classified as likely pathogenic/pathogenic by our lab (e.g., c.581C>G, p.Pro194Arg), supporting the critical relevance of codon 194 to ARSA protein function.
