Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_006329.4(FBLN5):c.716A>G (p.Glu239Gly), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces glutamic acid, which is acidic and polar, with glycine, which is neutral and non-polar, at codon 239 of the FBLN5 protein (p.Glu239Gly). This variant has not been reported in the literature in individuals affected with FBLN5-related conditions. This variant is present in population databases (no rsID available, gnomAD 0.006%). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt FBLN5 protein function. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
