Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000836.4(GRIN2D):c.896_899dup (p.Gly303fs), citing Invitae Variant Classification Sherloc (09022015): This sequence change creates a premature translational stop signal (p.Gly303Cysfs*14) in the GRIN2D gene. It is expected to result in an absent or disrupted protein product. However, the current clinical and genetic evidence is not sufficient to establish whether loss-of-function variants in GRIN2D cause disease. This variant is not present in population databases (gnomAD no frequency). This premature translational stop signal has been observed in individual(s) with clinical features of developmental and epileptic encephalopathy (Invitae). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
