Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001142800.2(EYS):c.9397_9398dup (p.Asn3133fs), citing Invitae Variant Classification Sherloc (09022015): This sequence change results in a frameshift in the EYS gene (p.Asn3133Lysfs*53). While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 12 amino acid(s) of the EYS protein and extend the protein by 40 additional amino acid residues. This variant is not present in population databases (gnomAD no frequency). This frameshift has been observed in individual(s) with inherited retinal dystrophy (PMID: 34662339). It has also been observed to segregate with disease in related individuals. This variant is also known as c.9460_9461dupAA (p.Asn3154Lysfs53). This variant disrupts a region of the EYS protein in which other variant(s) (c.9405T>A (p.Tyr3135*) ) have been determined to be pathogenic (PMID: 18976725, 30337596). This suggests that this is a clinically significant region of the protein, and that variants that disrupt it are likely to be disease-causing. For these reasons, this variant has been classified as Pathogenic.
