Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_020999.4(NEUROG3):c.320del (p.Arg107fs), citing Invitae Variant Classification Sherloc (09022015): This variant disrupts a region of the NEUROG3 protein in which other variant(s) (p.Arg107Ser) have been determined to be pathogenic (PMID: 16855267, 25901096, 27533310, 31178402, 31805014). This suggests that this is a clinically significant region of the protein, and that variants that disrupt it are likely to be disease-causing. For these reasons, this variant has been classified as Pathogenic. This variant has not been reported in the literature in individuals affected with NEUROG3-related conditions. This variant is not present in population databases (gnomAD no frequency). This sequence change creates a premature translational stop signal (p.Arg107Profs*29) in the NEUROG3 gene. While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 108 amino acid(s) of the NEUROG3 protein.
