Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000382.3(ALDH3A2):c.1349G>A (p.Trp450Ter), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the ALDH3A2 gene (transcript NM_000382.3) at coding-DNA position 1349, where G is replaced by A; at the protein level this means converts the codon for tryptophan at residue 450 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: This premature translational stop signal has been observed in individual(s) with Sjogren-Larsson syndrome (PMID: 34082469). This variant is not present in population databases (gnomAD no frequency). This sequence change creates a premature translational stop signal (p.Trp450*) in the ALDH3A2 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in ALDH3A2 are known to be pathogenic (PMID: 10577908, 10854114). For these reasons, this variant has been classified as Pathogenic. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site.
