Uncertain significance for GLUT1 deficiency syndrome 1, autosomal recessive — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_006516.4(SLC2A1):c.276-10C>G, citing Invitae Variant Classification Sherloc (09022015): This sequence change falls in intron 3 of the SLC2A1 gene. It does not directly change the encoded amino acid sequence of the SLC2A1 protein. This variant is present in population databases (rs746049837, gnomAD 0.007%). This variant has not been reported in the literature in individuals affected with SLC2A1-related conditions. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
