Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001042517.2(DIAPH3):c.2872A>T (p.Ile958Phe), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces isoleucine, which is neutral and non-polar, with phenylalanine, which is neutral and non-polar, at codon 958 of the DIAPH3 protein (p.Ile958Phe). This variant is present in population databases (rs761721396, gnomAD 0.01%). This variant has not been reported in the literature in individuals affected with DIAPH3-related conditions. An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be tolerated. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
