Pathogenic for Breast-ovarian cancer, familial, susceptibility to, 2 — the classification assigned by All of Us Research Program, National Institutes of Health to NM_000059.4(BRCA2):c.101_102insCCAATAAGTCTTA (p.Glu34delinsAspGlnTer), citing ACMG Guidelines, 2015. This variant lies in the BRCA2 gene (transcript NM_000059.4) at coding-DNA position 101 through coding-DNA position 102, inserting CCAATAAGTCTTA. Submitter rationale: This variant inserts 13 nucleotides in exon 3 of the BRCA2 gene, creating a frameshift and premature translation stop signal. This variant is expected to result in an absent or non-functional protein product. To our knowledge, this variant has not been reported in individuals affected with BRCA2-related disorders in the literature. This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). Loss of BRCA2 function is a known mechanism of disease (clinicalgenome.org). Based on the available evidence, this variant is classified as Pathogenic.

This study involves interpretation of variants in research participants for the purpose of population health screening. Participant phenotype was not available at the time of variant classification. Additional details can be found in publication PMID: 35346344, PMCID: PMC8962531
