Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_024334.3(TMEM43):c.682A>G (p.Ser228Gly), citing Ambry Variant Classification Scheme 2023: The p.S228G variant (also known as c.682A>G), located in coding exon 8 of the TMEM43 gene, results from an A to G substitution at nucleotide position 682. The serine at codon 228 is replaced by glycine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
