NM_000535.7(PMS2):c.391G>A (p.Val131Ile) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.V131I variant (also known as c.391G>A), located in coding exon 5 of the PMS2 gene, results from a G to A substitution at nucleotide position 391. The valine at codon 131 is replaced by isoleucine, an amino acid with highly similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this alteration remains unclear.
