Benign — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_004935.4(CDK5):c.312+3G>A, citing LabCorp Variant Classification Summary - May 2015: Variant summary: CDK5 c.312+3G>A alters a conserved nucleotide located close to a canonical splice site and therefore could affect mRNA splicing, leading to a significantly altered protein sequence. Consensus agreement among computation tools predict no significant impact on normal splicing. However, these predictions have yet to be confirmed by functional studies. The variant allele was found at a frequency of 0.00012 in 1610506 control chromosomes, predominantly at a frequency of 0.0051 within the Ashkenazi Jewish subpopulation in the gnomAD database, including 2 homozygotes. The observed variant frequency within Ashkenazi Jewish control individuals in the gnomAD database exceeds the estimated maximal expected allele frequency for disease-causing variants in CDK5. To our knowledge, no occurrence of c.312+3G>A in individuals affected with CDK5-related conditions and no experimental evidence demonstrating its impact on protein function have been reported.
