Uncertain significance for Autosomal dominant Parkinson disease 8 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_198578.4(LRRK2):c.4322G>C (p.Arg1441Pro), citing Invitae Variant Classification Sherloc (09022015): This variant is not present in population databases (gnomAD no frequency). This sequence change replaces arginine, which is basic and polar, with proline, which is neutral and non-polar, at codon 1441 of the LRRK2 protein (p.Arg1441Pro). This variant has not been reported in the literature in individuals affected with LRRK2-related conditions. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) has been performed at Invitae for this missense variant, however the output from this modeling did not meet the statistical confidence thresholds required to predict the impact of this variant on LRRK2 protein function. This variant disrupts the p.Arg1441 amino acid residue in LRRK2. Other variant(s) that disrupt this residue have been determined to be pathogenic (PMID: 15541309, 16750377, 17200152, 24565865). This suggests that this residue is clinically significant, and that variants that disrupt this residue are likely to be disease-causing. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
