Pathogenic for Autosomal recessive limb-girdle muscular dystrophy type 2A — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000070.3(CAPN3):c.63del (p.Pro22fs), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the CAPN3 gene (transcript NM_000070.3) at coding-DNA position 63, deleting one base; at the protein level this means shifts the reading frame starting at proline residue 22, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: For these reasons, this variant has been classified as Pathogenic. This premature translational stop signal has been observed in individual(s) with autosomal recessive limb-girdle muscular dystrophy (PMID: 10330340, 30919934). This variant is not present in population databases (gnomAD no frequency). This sequence change creates a premature translational stop signal (p.Pro22Glnfs*35) in the CAPN3 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in CAPN3 are known to be pathogenic (PMID: 10330340, 15689361).
