Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_024529.5(CDC73):c.1149C>G (p.Asp383Glu), citing Ambry Variant Classification Scheme 2023: The p.D383E variant (also known as c.1149C>G), located in coding exon 13 of the CDC73 gene, results from a C to G substitution at nucleotide position 1149. The aspartic acid at codon 383 is replaced by glutamic acid, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
