Pathogenic for Deficiency of galactokinase — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000154.2(GALK1):c.953_954del (p.Asp317_Tyr318insTer), citing Invitae Variant Classification Sherloc (09022015): This sequence change creates a premature translational stop signal (p.Tyr318*) in the GALK1 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in GALK1 are known to be pathogenic (PMID: 7670469, 10790206). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with GALK1-related conditions. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site. For these reasons, this variant has been classified as Pathogenic.
