NM_000038.6(APC):c.3700A>G (p.Ser1234Gly) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.S1234G variant (also known as c.3700A>G), located in coding exon 15 of the APC gene, results from an A to G substitution at nucleotide position 3700. The serine at codon 1234 is replaced by glycine, an amino acid with similar properties. This amino acid position is conserved. In addition, in silico predictors for this gene do not accurately predict pathogenicity. Based on the available evidence, the clinical significance of this alteration remains unclear.
