Pathogenic for Charcot-Marie-Tooth disease type 2 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_170707.4(LMNA):c.827A>C (p.Gln276Pro), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the LMNA gene (transcript NM_170707.4) at coding-DNA position 827, where A is replaced by C; at the protein level this means replaces glutamine at residue 276 with proline — a missense variant. Submitter rationale: This sequence change replaces glutamine, which is neutral and polar, with proline, which is neutral and non-polar, at codon 276 of the LMNA protein (p.Gln276Pro). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with autosomal dominant LMNA-related conditions (Invitae). In at least one individual the variant was observed to be de novo. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt LMNA protein function with a positive predictive value of 80%. For these reasons, this variant has been classified as Pathogenic.
