Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_001009999.3(KDM1A):c.301A>G (p.Ile101Val), citing Ambry Variant Classification Scheme 2023: The p.I101V variant (also known as c.301A>G), located in coding exon 1 of the KDM1A gene, results from an A to G substitution at nucleotide position 301. The isoleucine at codon 101 is replaced by valine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
