This variant disrupts a region of the KCNQ2 protein in which other variant(s) (p.Gly310Ser) have been determined to be pathogenic (PMID: 32942014, 35770094). This suggests that this is a clinically significant region of the protein, and that variants that disrupt it are likely to be disease-causing. This variant has not been reported in the literature in individuals affected with KCNQ2-related conditions. This variant is not present in population databases (gnomAD no frequency). This variant results in the deletion of part of exon 7 (c.928-7_933del) of the KCNQ2 gene. It is expected to disrupt RNA splicing. Variants that disrupt the donor or acceptor splice site typically lead to a loss of protein function (PMID: 16199547), and loss-of-function variants in KCNQ2 are known to be pathogenic (PMID: 14534157, 23692823, 27779742).
