This variant lies in the TPM1 gene (transcript NM_001018005.2) at the canonical splice donor site of the intron immediately after coding-DNA position 240, duplicating one base. Submitter rationale: This variant has not been reported in the literature in individuals affected with TPM1-related conditions. This variant is not present in population databases (gnomAD no frequency). This sequence change affects a donor splice site in intron 2 of the TPM1 gene It affects a nucleotide within the consensus splice site.
