Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_004311.4(ARL3):c.269A>C (p.Tyr90Ser), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces tyrosine, which is neutral and polar, with serine, which is neutral and polar, at codon 90 of the ARL3 protein (p.Tyr90Ser). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with autosomal dominant retinitis pigmentosa (Invitae). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive. This variant disrupts the p.Tyr90Cys amino acid residue in ARL3. Other variant(s) that disrupt this residue have been determined to be pathogenic (PMID: 26964041, 30932721). This suggests that this residue is clinically significant, and that variants that disrupt this residue are likely to be disease-causing. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
