Submitter rationale: The p.I3008M variant (also known as c.9024T>G), located in coding exon 22 of the BRCA2 gene, results from a T to G substitution at nucleotide position 9024. The isoleucine at codon 3008 is replaced by methionine, an amino acid with highly similar properties. The results from two saturation genome editing-based studies, including a haploid cell-survival assay and a humanized mouse embryonic stem cell line assay of drug response and survival, are discordant for this nucleotide substitution (Huang H et al.
