Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_015047.3(EMC1):c.1859A>C (p.Lys620Thr), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces lysine, which is basic and polar, with threonine, which is neutral and polar, at codon 620 of the EMC1 protein (p.Lys620Thr). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with EMC1-related conditions. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt EMC1 protein function.
