Pathogenic for Congenital muscular hypertrophy-cerebral syndrome — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_006306.4(SMC1A):c.909G>C (p.Glu303Asp), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the SMC1A gene (transcript NM_006306.4) at coding-DNA position 909, where G is replaced by C; at the protein level this means replaces glutamic acid at residue 303 with aspartic acid — a missense variant. Submitter rationale: For these reasons, this variant has been classified as Pathogenic. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt SMC1A protein function. This missense change has been observed in individual(s) with Cornelia de Lange (Invitae). In at least one individual the variant was observed to be de novo. This variant is not present in population databases (gnomAD no frequency). This sequence change replaces glutamic acid, which is acidic and polar, with aspartic acid, which is acidic and polar, at codon 303 of the SMC1A protein (p.Glu303Asp).
