Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_004304.5(ALK):c.3668T>C (p.Met1223Thr), citing Ambry Variant Classification Scheme 2023: The p.M1223T variant (also known as c.3668T>C), located in coding exon 24 of the ALK gene, results from a T to C substitution at nucleotide position 3668. The methionine at codon 1223 is replaced by threonine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
