Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_005739.4(RASGRP1):c.945dup (p.Val316fs), citing Invitae Variant Classification Sherloc (09022015): For these reasons, this variant has been classified as Pathogenic. This variant has not been reported in the literature in individuals affected with RASGRP1-related conditions. This variant is not present in population databases (gnomAD no frequency). This sequence change creates a premature translational stop signal (p.Val316Cysfs*4) in the RASGRP1 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in RASGRP1 are known to be pathogenic (PMID: 11017103, 27776107, 28822832).
