Uncertain significance for Primary ciliary dyskinesia — the classification assigned by Ambry Genetics to NM_001277115.2(DNAH11):c.1643A>G (p.Asp548Gly), citing Ambry Variant Classification Scheme 2023: The p.D548G variant (also known as c.1643A>G), located in coding exon 9 of the DNAH11 gene, results from an A to G substitution at nucleotide position 1643. The aspartic acid at codon 548 is replaced by glycine, an amino acid with similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
