Uncertain significance for Cardiovascular phenotype; Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_001042492.3(NF1):c.2923C>T (p.His975Tyr), citing Ambry Variant Classification Scheme 2023: The p.H975Y variant (also known as c.2923C>T), located in coding exon 22 of the NF1 gene, results from a C to T substitution at nucleotide position 2923. The histidine at codon 975 is replaced by tyrosine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
