Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_002048.3(GAS1):c.396G>T (p.Gln132His), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces glutamine, which is neutral and polar, with histidine, which is basic and polar, at codon 132 of the GAS1 protein (p.Gln132His). This variant is present in population databases (rs759982942, gnomAD 0.09%), and has an allele count higher than expected for a pathogenic variant. This variant has not been reported in the literature in individuals affected with GAS1-related conditions. ClinVar contains an entry for this variant (Variation ID: 2738413). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be tolerated.
