NM_020975.6(RET):c.803C>G (p.Ser268Trp) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.S268W variant (also known as c.803C>G), located in coding exon 4 of the RET gene, results from a C to G substitution at nucleotide position 803. The serine at codon 268 is replaced by tryptophan, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
