Therefore, it has been classified as a Variant of Uncertain Significance. Experimental studies have shown that this missense change affects DCX function (PMID: 30979500). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive. This missense change has been observed in individual(s) with DCX-related conditions (PMID: 20726879). It has also been observed to segregate with disease in related individuals. This variant is not present in population databases (gnomAD no frequency). This sequence change replaces glutamic acid, which is acidic and polar, with lysine, which is basic and polar, at codon 2 of the DCX protein (p.Glu2Lys).
