Pathogenic for Metachromatic leukodystrophy — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000487.6(ARSA):c.629T>C (p.Leu210Pro), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the ARSA gene (transcript NM_000487.6) at coding-DNA position 629, where T is replaced by C; at the protein level this means replaces leucine at residue 210 with proline — a missense variant. Submitter rationale: For these reasons, this variant has been classified as Pathogenic. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt ARSA protein function. This variant is also known as c.371T>C (p.Leu124Pro). This missense change has been observed in individual(s) with metachromatic leukodystrophy (PMID: 27779215, 30057904). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. This variant is not present in population databases (gnomAD no frequency). This sequence change replaces leucine, which is neutral and non-polar, with proline, which is neutral and non-polar, at codon 210 of the ARSA protein (p.Leu210Pro).
