Likely pathogenic for Neurofibromatosis, type 2 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000268.4(NF2):c.676-7T>G, citing Invitae Variant Classification Sherloc (09022015): This sequence change falls in intron 7 of the NF2 gene. It does not directly change the encoded amino acid sequence of the NF2 protein. RNA analysis indicates that this variant induces altered splicing and likely results in a shortened protein product. This variant is not present in population databases (gnomAD no frequency). This variant has been observed in individual(s) with clinical features of neurofibromatosis type 2 (PMID: 15684865; internal data). It has also been observed to segregate with disease in related individuals. ClinVar contains an entry for this variant (Variation ID: 2737029). Studies have shown that this variant results in skipping of skipping of exon 8 , but is expected to preserve the integrity of the reading-frame (internal data).
