Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001145809.2(MYH14):c.1609G>A (p.Asp537Asn), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the MYH14 gene (transcript NM_001145809.2) at coding-DNA position 1609, where G is replaced by A; at the protein level this means replaces aspartic acid at residue 537 with asparagine — a missense variant. Submitter rationale: This sequence change replaces aspartic acid, which is acidic and polar, with asparagine, which is neutral and polar, at codon 529 of the MYH14 protein (p.Asp529Asn). This variant is present in population databases (no rsID available, gnomAD 0.004%). This missense change has been observed in individual(s) with autosomal dominant nonsyndromic deafness (PMID: 25719458). In at least one individual the variant was observed to be de novo. This variant is also known as c.1609G>A (p.D537N). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) has been performed at Invitae for this missense variant, however the output from this modeling did not meet the statistical confidence thresholds required to predict the impact of this variant on MYH14 protein function. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
