This variant lies in the UNC13D gene (transcript NM_199242.3) at coding-DNA position 1387, where C is replaced by T; at the protein level this means converts the codon for glutamine at residue 463 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: This premature translational stop signal has been observed in individual(s) with familial hemophagocytic lymphohistiocytosis (PMID: 18492689). For these reasons, this variant has been classified as Pathogenic. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site.
